    A management study was conducted to specify activities and problems encountered during the development of procedures for documentation and crew training on experiments, as well as during the design, integration, and delivery of a life sciences experiment payload to Johnson Space Center for a 7 day simulation of a Spacelab mission. Conclusions and recommendations to project management for current and future Ames' life sciences projects are included. Broader issues relevant to the conduct of future scientific missions under the constraints imposed by the environment of space are also addressed

    Spectral statistics in chaotic systems with a point interaction

    We consider quantum systems with a chaotic classical limit that are perturbed by a point-like scatterer. The spectral form factor K(tau) for these systems is evaluated semiclassically in terms of periodic and diffractive orbits. It is shown for order tau^2 and tau^3 that off-diagonal contributions to the form factor which involve diffractive orbits cancel exactly the diagonal contributions from diffractive orbits, implying that the perturbation by the scatterer does not change the spectral statistic.     Intensity distribution in rotational line spectra

    Completely resolved Doppler-free rotational line spectra of six vibronic two-photon bands in benzene C6 H6 and C6 D6 are presented. The excited final states possess different excess energies in S1 (1567 to 2727 cm−1 ) and are embedded in dense manifolds of background states with differing densities of states (1<rho<60 1/cm−1 ). The bands are analyzed by a statistical procedure. The intensity distribution of several hundreds of lines of each band is investigated. It is found that all weakly perturbed bands display a similar, peaked intensity distribution while in strongly perturbed bands the number of lines decreases monotonically with increasing intensity. We consider the set of C1C^1 maps f:MMf:M\to M which have no absolutely continuous (with respect to Lebesgue) invariant probability measure. We show that this is a residual (dense Gδ)setintheG_\delta) set in the C^1$ topology. In the course of the proof, we need a generalization of the usual Rokhlin tower lemma to non-invariant measures.     To learn how ground-based personnel of a space project plan and organize their work and how such planning and organizing relate to work outcomes, longitudinal study of the management and execution of the Space Lab Mission Development Test 3 (SMD 3) was performed at NASA Ames Research Center. A view of the problems likely to arise in organizations and some methods of coping with these problems are presented as well as the conclusions and recommendations that pertain strictly to SMD 3 management. Emphasis is placed on the broader context of future space facility projects and additional problems that may be anticipated. A model of management that may be used to facilitate problem solving and communication - management by objectives (MBO) is presented. Some problems of communication and emotion management that MBO does not address directly are considered. Models for promoting mature, constructive and satisfying emotional relationships among group members are discussed

    Geometrical theory of diffraction and spectral statistics

    We investigate the influence of diffraction on the statistics of energy levels in quantum systems with a chaotic classical limit. By applying the geometrical theory of diffraction we show that diffraction on singularities of the potential can lead to modifications in semiclassical approximations for spectral statistics that persist in the semiclassical limit 0\hbar \to 0. This result is obtained by deriving a classical sum rule for trajectories that connect two points in coordinate space.Comment: 14 pages, no figure, to appear in J. Phys.

    Effects of pre-hydration on hydraulic properties on Portland cement and synthetic clinker phases

    The ageing of cements (CEM I 52,5 R and API Oilwell Class G) and also the pure cement clinker minerals (C3S, pure C3A, Na2O-doped C3A, C4AF and calcium sulphate hemihydrate) has been studied at 60% and 85% RH using in-situ XRD, calorimetry, ESEM, XPS and FTIR-ATR. This combination of techniques has identified the key components involved in the surface pre-hydration process. Pre-hydration of cement leads to retardation of hydration. In-situ XRD shows the retardation of Portlandite formation after prehydration, which explains the heat development seen via calorimetry. XPS and ESEM investigations show surface modification after prehydration for C3S, C3A and doped C3A (4% Na2O). Pre-hydration of cement leads to modification of the surface area and surface charge, which can strongly influence the reactivity of bulk cements and their interaction with admixtures. This is relevant to the quality of cement during storage

    A protocol for registration and correction of multicolour STED superresolution images.

    Multicolour fluorescence imaging by STimulated Emission Depletion (STED) superresolution microscopy with doughnut-shaped STED laser beams based on different wavelengths for each colour channel requires precise image registration. This is especially important when STED imaging is used for colocalisation studies of two or more native proteins in biological specimens to analyse nanometric subcellular spatial arrangements. We developed a robust postprocessing image registration protocol, with the aim to verify and ultimately optimise multicolour STED image quality. Importantly, this protocol will support any subsequent quantitative localisation analysis at nanometric scales. Henceforth, using an approach that registers each colour channel present during STED imaging individually, this protocol reliably corrects for optical aberrations and inadvertent sample drift. To achieve the latter goal, the protocol combines the experimental sample information, from corresponding STED and confocal images using the same optical beam path and setup, with that of an independent calibration sample. As a result, image registration is based on a strategy that maximises the cross-correlation between sequentially acquired images of the experimental sample, which are strategically combined by the protocol. We demonstrate the general applicability of the image registration protocol by co-staining of the ryanodine receptor calcium release channel in primary mouse cardiomyocytes. To validate this new approach, we identify user-friendly criteria, which - if fulfilled - support optimal image registration. In summary, we introduce a new method for image registration and rationally based postprocessing steps through a highly standardised protocol for multicolour STED imaging, which directly supports the reproducibility of protein co-localisation analyses. Although the reference protocol is discussed exemplarily for two-colour STED imaging, it can be readily expanded to three or more colours and STED channels
